ABSTRACT

Introducción: El Campamento Tortuguero de Cedeño ha sido el sitio menos investigado del Golfo de Fonseca, donde se protege a la tortuga golfina en Honduras desde 1975. Objetivo: Evaluar la anidación de la tortuga Golfina (Lepidochelys olivacea) durante la temporada de veda entre el 2011 y 2021 en Campamento Tortuguero Cedeño, Choluteca, Honduras. Métodos: Entre 2011 a 2021, se llevó a cabo el monitoreo diario de las actividades de anidación durante la veda del 1 al 25 de septiembre. Los patrullajes se realizaron entre las 6:00-18:00 h, y las 18:00-5:00 h. Se registró el número total de tortugas que anidaban y se recogieron sus huevos, que se transportaron al criadero, donde se tabularon los resultados de las puestas y las crías. Resultados: Se registró un total 1 065 tortugas de L. olivacea, 95 051 huevos recolectados, 1 065 nidos marcados en tres playas que fueron reubicados en viveros artificiales y una eclosión exitosa de 62 747 neonatos. La playa Las Doradas fue el sitio con el mayor número de tortugas anidadoras, seguido de Los Delgaditos y por último Cedeño. El promedio de la frecuencia de anidación fue de 96 nidos. Del 2011 al 2021 el esfuerzo de recolección de los nidos aumentó en un 91.6 %, pasando de 84 a 161 nidos. El número de personas patrullando se asoció con la cantidad de nidos detectados en las playas. Conclusión: Los esfuerzos de monitoreo y conservación para la especie han indicado que ha habido un incremento en la anidación de L. olivacea en las tres playas, con un mayor incremento en Playa Las Doradas. Este escenario comprueba la funcionalidad de la veda en esta zona.


Introduction: The Cedeño Turtle Camp has been the least researched site in the Fonseca Gulf, where Olive Ridley Turtles in Honduras have been protected since 1975. Objective: To evaluate the nesting of Olive Ridley turtles (Lepidochelys olivacea) during the closed season from 2011 to 2021 in Campamento Tortuguero Cedeño, Choluteca, Honduras. Methods: From 2011 to 2021, daily monitoring of nesting activities was conducted during the closed season from the 1st to 25th of September. Patrols were conducted between 6:00-18:00 h, and 18:00-5:00 h. The total number of nesting turtles was recorded, and their eggs were collected and transported to the hatchery, where clutch and hatchling performance were tabulated. Results: A total of 1 065 L. olivacea turtles were recorded, 95 051 eggs collected, 1 065 nests marked on three beaches that were relocated in artificial hatcheries and a successful hatching of 62 747 hatchlings. Las Doradas beach was the site with the highest number of nesting turtles, followed by Los Delgaditos and lastly Cedeño. The average nesting frequency was 96 nests. From 2011 to 2021 the nest collection effort increased by 91.6 %, from 84 to 161 nests. The number of people patrolling was associated with the number of nests detected on the beaches. Conclusion: Monitoring and conservation efforts for L. olivacea in the Campamento Tortuguero Cedeño show a positive trend in nesting with a greater increase in Playa Las Doradas. This scenario proves the functionality of the closed season in this area.

ABSTRACT

Nos proponemos compartir resultados de una investigación que tuvo como objetivo estudiar los procesos de gestión colectiva en dos emprendimientos laborales autogestivos de Uruguay, que trabajan desde la perspectiva de la desmanicomialización. Se trató de una investigación cualitativa, situada y experiencial a partir del despliegue de una etnografía. En este artículo abordaremos los resultados de dos objetivos específicos, uno sobre las estrategias y metodologías universitarias vinculadas a los procesos de conformación y sostén de dichas experiencias y otro sobre las estrategias desplegadas para acompañar el padecimiento de lxs integrantes. Compartiremos algunos resultados en relación a las preguntas: ¿para qué y por qué se acompaña? ¿qué es lo que se acompaña? ¿cómo se acompaña?. Identificamos que ambos proyectos se llevan adelante desde la extensión universitaria, a partir de un proceso de incubación de emprendimientos cooperativos integrado solo por personas usuarias de servicios de salud mental, que denominamos: autogestión-solo-de-ellos. Este proceso implicó acompañar y sostener un doble pasaje, o sea generar ruptura con la lógica manicomial y con la lógica salarial para configurar y consolidar un emprendimiento de gestión colectiva. Esto significó para los equipos universitarios acompañar y desplegar distintas estrategias en relación a la triada: trabajo-acogida-vida. Destacamos tres componentes transversales del acompañamiento que fueron: no sustituir (hacer con), sostener otras temporalidades y abordaje interdisciplinario y en red.Durante el proceso de incubación se produce una metamorfosis a partir de que egresadxs universitarixs deciden integrar las experiencias como socixs/cooperativistas e inician un proceso que definimos como: autogestión-de-un-nosotrxs.


Propomos compartilhar resultados de uma pesquisa que teve como objetivo estudar os processos de gestão coletiva em duas empresas trabalhistas autogeridas no Uruguai, que funcionam na perspectiva da desmanicomalização. Tratou-se de uma investigação qualitativa, situada e experiencial baseada no desdobramento de uma etnografia. Neste artigo abordaremos os resultados de dois objetivos específicos, um sobre as estratégias e metodologias universitárias vinculadas aos processos de formação e sustentação dessas experiências e outro sobre as estratégias utilizadas para acompanhar o sofrimento dos membros. Compartilharemos alguns resultados em relação às questões: para que e por que é acompanhado? O que é acompanhado? E como é acompanhado? Identificamos que ambos os projetos são realizados pela extensão universitária, a partir de um processo de incubação de empreendimentos cooperativos formados apenas por usuários de serviços de saúde mental, o que denominamos: autogestão-só-deles. Este processo envolveu acompanhar e sustentar uma dupla passagem, ou seja, gerar uma ruptura com a lógica asilar e com a lógica salarial para configurar e consolidar um empreendimento de gestão coletiva. Isso significou que as equipes universitárias acompanhassem e implantassem diferentes estratégias em relação à tríade: trabalho-acolhimento-vida. Destacamos três componentes transversais do apoio, que foram: não substituir (fazer com), apoiar outras temporalidades e uma abordagem interdisciplinar e em rede. Durante o processo de incubação, ocorre uma metamorfose quando os graduados universitários decidem integrar as experiências como parceiros/cooperativistas e iniciam um processo que definimos como: autogestão-de-um-nós.


We propose to share results of a research that aimed to study the collective management processes in two self-managed labor enterprises in Uruguay, which work from the perspective of demanicomalization. It was a qualitative, situated and experiential investigation based on the deployment of an ethnography. In this article, we will address the results of two specific objectives, one on the university strategies and methodologies linked to the processes of formation and support of these experiences and another on the strategies deployed to accompany the suffering of the members. We will share some results in relation to the questions: why and what for is it accompanied? What is accompanied? And how is it accompanied? We identified that both projects are carried out by the university extension, based on an incubation process of cooperative ventures made up only of users of mental health services, which we call: self-management-only-of-them. This process involved accompanying and sustaining a double passage; that is, generating a break with the asylum logic and with the salary logic to configure and consolidate a collective management undertaking. This meant for the university teams to accompany and deploy different strategies in relation to the triad: work-welcome-life. We highlighted three transversal components of support, which were not substituting (doing with), supporting other temporalities, and an interdisciplinary and network approach. During the incubation process, a metamorphosis occurs when university graduates decide to integrate the experiences as partners/cooperativists and begin a process that we define as: self-management-of-an-us.

ABSTRACT

Gynerium sagittatum es una gramínea ampliamente utilizada en la costa Caribe colombiana como fuente de fibra natural para la elaboración de artesanías, particularmente por la comunidad Zenú. En la presente investigación se evaluó el efecto de diferentes concentraciones de enzimas: celulasa y macerozima a diferentes tiempos de incubación y sus interacciones en el aislamiento de protoplastos. Los protoplastos se obtuvieron del mesófilo foliar de vitroplantas de G. sagittatum expuesto a combinaciones enzimáticas de celulasa (1.5 y 2.0%), con macerozima (0.3, 0.6 y 0.9%), durante 3, 6 y 9 horas de incubación, para un total de 18 tratamientos con 5 réplicas cada uno. Los mayores números de protoplastos aislados correspondieron a T18 (2.0% celulasa, 0.9% macerozima), T12 (2.0% de celulasa, 0.3% macerozima), T3 (1.5% de celulasa, 0.3% de macerozima) y T6 (1.5% de celulasa, 0.6% de macerozima) por 9 horas de incubación cada uno, con valores de 88.625, 83.000, 75.000 y 53.375 protoplastos/mL respectivamente. El tiempo de incubación fue significativo en el aislamiento de los protoplastos (p<0.05). Las predicciones entre factores mostraron que una interacción de 2.0% de celulasa y 0.9% de macerozima permite obtener 44.302 protoplastos/mL, mientras que las interaciciones tiempo de incubación-celulasa y tiempo de incubación-macerozima mostraron que es posible obtener 72.073 y 71.212 protoplastos/mL con 2.0% de celulasa y 0.9% macerozima por 9 horas de incubación cada una respectivamente. Los resultados indican que la aplicación de estas enzimas permite obtener cantidades considerables de protoplastos de G. sagittatum a partir de explantes cultivados in vitro.


Gynerium sagittatum is a graminaceous plant widely used in the Caribbean coast of Colombia as a natural fiber source for the elaboration of handicrafts, particularly by the Zenú community. In the present investigation, the effect of different concentrations of cellulase and macerozyme enzymes at different incubation times and their interaction in the isolation of protoplasts was evaluated. Protoplasts were obtained from leaf mesophyll of G. sagittatum vitroplants exposed to enzymatic combinations of cellulase (1.5 and 2.0%), with macerozyme (0.3, 0.6 and 0.9%), for 3, 6 and 9 hours of incubation, for a total of 18 treatments with 5 replicates each. The highest numbers of isolated protoplasts corresponded to T18 (2.0% cellulase, 0.9% macerozyme), T12 (2.0% cellulase, 0.3% macerozyme), T3 (1.5% cellulase, 0.3% macerozyme) and T6 (1.5% cellulase, 0.6% macerozyme); at 9 hours incubation. The protoplast number for these treatments were: 88.625, 83.000, 75.000 and 53.375 protoplasts/mL respectively. Incubation time was significant in the isolation of protoplasts (p<0.05). The predictions between the factors showed that with an interaction of 2.0% cellulase and 0.9% macerozyme it is possible to obtain 44.302 protoplasts/mL, likewise, the incubation time-cellulase and incubation time-macerozyme interactions showed that it is possible to obtain 72.073 and 71.212 protoplasts/mL with 2.0% cellulase and 0.9% macerozyme for 9 hours of incubation respectively. The results indicate that the use of these enzymes and time, allows the isolation of of protoplasts from G. sagittatum in vitro plants.

ABSTRACT

SUMMARY: The domestic chicken is a species of bird that has been extensively studied in regard to its biology and as a model organism for science. The reproduction of the species is by the laying of fertilized eggs, which in a period of 21 days will develop a chick inside. Several methods have been described to develop embryos ex-ovo, allowing the observation and manipulation of the organism. This work has the propose to standardize a method that allows the development of the embryos inside the artificial incubation system, which has a low cost and is easy to make. In this work, 100 chicken eggs were used to study the effects of humidity, mineral supplementation, and the preincubation time of the egg on the incubation ex-ovo of the embryos. Embryo development was documented through the different days. Pulverized eggshell was selected as an optimal source to provide calcium, magnesium, phosphorus, and other minerals to the developing embryo. By providing 900-1200 mg of pulverized eggshell, 40 mL of the 0.001 % solution of benzalkonium chloride, and a preincubation time of approximately 56 h, the embryos were able to develop until 19 days, and even though they did not reach hatching, the incubation conditions that allowed the survival and development of embryos until late stages were achieved. Thus, due to the conditions established for calcium, humidity and preincubation time, in the present work, the chicks reached 19 days of development.


El pollo doméstico es una especie de ave que ha sido ampliamente estudiada en cuanto a su biología y como organismo modelo para la ciencia. La reproducción de la especie es por la puesta de huevos fecundados, que en un período de 21 días desarrollarán un polluelo en su interior. Se han descrito varios métodos para desarrollar embriones ex-ovo, permitiendo la observación y manipulación del organismo. Este trabajo tuvo como objetivo estandarizar un método que permita el desarrollo de los embriones dentro del sistema de incubación artificial, el cual tiene un bajo costo y es fácil de realizar. En este trabajo se utilizaron 100 huevos de gallina para estudiar los efectos de la humedad, la suplementación mineral y el tiempo de preincubación del huevo sobre la incubación ex-ovo de los embriones. El desarrollo embrionario se documentó a través de los diferentes días. Se seleccionó la cáscara de huevo pulverizada como una fuente óptima para proporcionar calcio, magnesio, fósforo y otros minerales al embrión en desarrollo. Al suministrar 900-1200 mg de cáscara de huevo pulverizada, 40 mL de la solución de cloruro de benzalconio al 0.001 % y un tiempo de preincubación de aproximadamente 56 h, los embriones lograron desarrollarse hasta los 19 días, y aunque no llegaron a eclosionar, los embriones lograron desarrollarse hasta los 19 días. Se lograron condiciones de incubación que permitieron la supervivencia y desarrollo de los embriones hasta etapas tardías. Así, debido a las condiciones establecidas de calcio, humedad y tiempo de preincubación, en el presente trabajo los pollitos alcanzaron los 19 días de desarrollo.

Introducción. Psarocolius decumanus es una especie relativamente abundante en el departamento de Sucre, Colombia, pero no se cuenta con información suficiente acerca de su reproducción. Materiales y Métodos. A través de observación directa y recorridos libres se determinó el número de nidos por colonias, los árboles donde son construidos, materiales utilizados, la altura a que se encontraban y se obtuvo información sobre la duración del periodo reproductivo y comportamiento parental, durante varios meses de muestreo (febrero a octubre), en diferentes localidades de la región. Resultados. Los nidos se construyen en colonias de hasta diez nidos y son defendidos por individuos de ambos sexos, aunque en su construcción, alimentación de los pichones y protección de cada nido solo intervienen las hembras. La ubicación de los nidos en árboles altos, aislados, en zonas abiertas o semiabiertas, en el extremo de ramas finas, parece constituir una estrategia anti-depredación. La construcción comienza en febrero, la puesta de los huevos es a mediados de marzo, la incubación dura 18 días aproximadamente y los pichones independientes fueron observados 33 días después de la eclosión de los huevos, a finales de mayo y principios de junio. Este proceso debe extenderse varios meses, ya que se observó construcción de nidos hasta junio. Conclusión. Casi todas las características de la anidación en esta especie están enfocadas, sobre todo, a la protección contra la depredación y el parasitismo.


Introduction. Psarocolius decumanus is a relatively abundant species in the department of Sucre, Colombia and there is not enough information about its reproduction. Materials and methods. Through direct observation and free routes, the number of nests per colonies was determined, also the trees where they were built, the materials used, the height they were at, and information was obtained on the duration of the reproductive period and parental behavior, for several months of sampling (February to October) in different localities of the region. Results. The nests are built in colonies of up to 10 nests and are defended by individuals of both sexes, although in its construction, feeding the pigeons and protection of each single nest, involved only females. The location of nests in tall, isolated trees, in open or semi-open areas, at the end of thin branches, seems to be an anti-predation strategy. The construction begins in February, the eggs are laid in mid-March, the incubation lasts approximately eighteen days and the independent chicks were observed thirty-three days after the hatching of the eggs, at the end of May and the beginning of June. This process should extend several months, since construction of nests were observed until June. Conclusion. Almost all nesting characteristics in this species are focused, above all, on protection against predation and parasitism.

ABSTRACT

Objective:To investigate the effect of different incubation time of aminolevulinic acid (ALA) on photodynamic inhibition of Propionibacterium acnes biofilms. Methods:Propionibacterium acnes biofilms were formed in 24-well plates with pre-placed cell slides and 96-well plates. The formation of the biofilm structure was observed by confocal laser scanning microscopy (CLSM) , and the growth activity of the biofilm was assessed by the tetrazolium salt XTT assay. The in vitro successfully constructed biofilm models were divided into 6 groups: negative control group receiving neither ALA treatment nor LED radiation, ALA group incubated with ALA alone for 30 minutes, LED group receiving LED radiation alone, ALA-PDT1 group, ALA-PDT2 group and ALA-PDT3 group incubated with ALA for 15, 30 and 60 minutes respectively followed by LED radiation. After the treatment, CLSM was performed to observe the biofilm structure, as well as to determine the dead/living bacteria ratio, and XTT assay to assess the growth activity of the biofilm. Differences among groups were analyzed using one-way analysis of variance and least significant difference- t test. Results:CLSM showed that the Propionibacterium acnes biofilm model was successfully constructed in vitro. The dead/living bacteria ratios were 0.90 ± 0.16, 1.75 ± 0.19, and 2.57 ± 0.32 in the ALA-PDT1 group, ALA-PDT2 group and ALA-PDT3 group respectively, which were significantly higher than the dead/living bacteria ratio in the negative control group (0.31 ± 0.01; t= 55.56, 138.62, 74.64, respectively, all P<0.001) ; the biofilm viability value was significantly lower in the ALA-PDT1 group, ALA-PDT2 group and ALA-PDT3 group (0.35 ± 0.02, 0.26 ± 0.02, 0.18 ± 0.01, respectively) than in the negative control group (0.43 ± 0.00; t= 35.66, 2.64, 110.96, respectively, all P < 0.001) . CLSM showed that the structure of the Propionibacterium acnes biofilm was destroyed under the action of ALA-PDT, and the destruction was aggravated with the prolongation of incubation time of ALA. Conclusion:The prolongation of incubation time of ALA can enhance the inhibitory effect of ALA-PDT on Propionibacterium acnes biofilms.

Objective:The article aims to explore the positive promoting effect of the construction of a medical university’s incubation platform on the Industry-University-Institute cooperation and biomedical scientific and technological achievements transformation.Methods:Through literature review, the article studied the existing mode of Industry-University-Institute cooperation in universities, analyzed the main bottlenecks, and summarized the practical exploration experience of ″Shanghai Pharmaceuticals-SJTUSM Innovative Achievements″ incubation platform.Results:The incubation platform effectively promoted the process and system construction of the scientific and technological achievements transformation in the university, and improved the project mining level and platform support function.Conclusions:The practical exploration of the incubation platform lays a foundation for the construction of the biomedical industrial park. Through the in-depth construction of the " Government-Industry-University-Institute-Investment-Inventor" development chain, the platform can help to promote strategic innovation and industrialization of achievements, making the " first-in-class" medicine in China.

ABSTRACT

Objective:To explore solutions to the "grey zone" of activated partial thromboplastin time (APTT) mixing study, and establish the clinical application pathway of it.Methods:Patients treated in West China Hospital of Sichuan University from January 1, 2018, to December 31, 2019, with a prolonged APTT were included in this study. The ROC curve was used to analyze the"cut-off"of different methods and explore solutions to the "grey zone" by combination of the 1∶1 and 4∶1 mixing study. Similar samples from January 1, 2020 to December 31, 2020 were included to verify the diagnostic efficiency of the clinical application pathway.Results:The traditional Rosner index criterion had a low diagnostic accuracy in differentiating factor deficiencies from inhibitors. A total of 49 cases (15%) in the establishment group and validation group were located in the "grey zone". The optimal cut-off value of the Rosner index in our 1∶1 mixing study for determining factor deficiency was 5.0%, and inhibitor was 9.1%. The sample between 5.0% and 9.0% needed 4∶1 mixing studies, which could significantly improve the detection sensitivity of inhibitors. The percentage of extended time after incubation-P (1∶1 mixing>10.8% and 4∶1 mixing>13.5%) was better than the traditional criterion mentioned by"consensus"in determining whether the inhibitor was time-dependent. The sensitivity, specificity, positive predictive value and negative predictive value of combined the 1∶1 and 4∶1 mixing study in differentiating factor deficiencies from inhibitors all attained more than 90%. Only 7% (3/43)of inhibitors were incorrectly classified into the factor deficiency group by the combination, which was 20.9% (9/43) by traditional criterion. The specificity for detecting time-dependent inhibitor was increased from 54.2% to 100%, and accuracy was increased from 63.3% to 97.4%.Conclusions:The combination of 1∶1 and 4∶1 mixing study can better resolve the "grey zone". The established clinical application pathway is beneficial for the further promotion and clinical application of APTT mixing study.

Resumen Introducción: Para incrementar el número de crías de tortugas marinas, es necesario mejorar estrategias de conservación, como incubación de nidos, en condiciones de vivero que contrarresten los efectos de las condiciones climáticas extremas. Objetivo: Comparar cinco parámetros de éxito reproductivo (éxito de eclosión, huevos sin desarrollo aparente, crías muertas, mortalidad embrionaria y período de incubación) de la tortuga marina Lepidochelys olivacea. Métodos: Se evaluó la incubación de nidos bajo dos períodos de reubicación de nidos (P1 ~28.8 °C agosto-septiembre y P2 ~27.1 °C octubre-noviembre, temperatura ambiental promedio, bajo condiciones de vivero, en el 2018 en Guerrero, México. Resultados: Se encontraron diferencias significativas en el éxito de eclosión y la mortalidad embrionaria entre los períodos de reubicación de nidos (P < 0.001). En P1, se observaron valores más bajos de éxito de eclosión (77.0 % P1 vs 88.6 % P2) y valores más altos de mortalidad embrionaria (13.7 % P1 vs 3.3 % P2) comparados con los de P2. Es importante señalar que, en el presente estudio la mortalidad embrionaria se presentó en una etapa tardía de desarrollo, es decir, en embriones casi completamente desarrollados. No se encontraron diferencias entre periodos respecto a los parámetros de huevos sin desarrollo aparente, crías muertas y período de incubación. Conclusiones: Las diferencias entre las condiciones ambientales durante el primer período de reubicación de nidos en comparación con el segundo período parecen afectar el éxito de eclosión y mortalidad embrionaria de L. olivacea. Por lo tanto, es importante tomar medidas al respecto durante esta etapa de incubación para tratar de mejorar la incubación de nidos bajo condiciones de vivero.


Abstract Introduction: In order to increase the number of sea turtle hatchlings, it is necessary to improve conservation strategies, such as nest incubation in hatchery conditions that counteract the effects of extreme climatic conditions. Objective: To compare five reproductive success parameters (hatching success, eggs with no apparent embryonic development, dead hatchlings, embryonic mortality, and incubation period) of the sea turtle Lepidochelys olivacea. Methods: The incubation of nests was evaluated under two periods of nest relocation, P1 ~28.8 °C (August and September) and P2 ~27.1 °C (October and November), average ambient temperature, under hatchery condition, in 2018 in Guerrero, Mexico. Results: Significant differences were found in hatching success and embryonic mortality between nest relocation periods (P < 0.001). In P1, lower values of hatching success (77.0 % P1 vs 88.6 % P2) and higher values of embryonic mortality (13.7 % P1 vs 3.3 % P2) were observed compared to those of P2. It's important pointing that, in the present study, embryonic mortality occurred at a late stage of development, that is, in almost fully developed embryos. No differences were found between periods regarding the parameters eggs with no apparent embryonic development, dead hatchlings and incubation period. Conclusions: The differences between the environmental conditions during the first period of nest relocation compared to the second period, respectively, seem to affect the hatching success and embryonic mortality of L. olivacea. Therefore, it is important to take action on this issue during this incubation stage to try to improve the incubation of nests under hatchery conditions.

ABSTRACT

This study aimed to evaluate the effectiveness of ultraviolet light in reducing bacterial load of eggshells and the impact of experimental disinfection on hatching, embryo mortality, and time-borne distribution using broiler breeder hens of different ages (38, 42, and 48 weeks old). Fertile eggs were subjected to different exposure periods (5, 7, and 9 minutes) of UV light (UV-C) with a 254 nm wavelength. For controls, eggs disinfected with paraformaldehyde (5.3 g/m3) and eggs not disinfected (NC). After subjection to disinfection protocols, the eggs were placed into sterile plastic bags containing 20 mL of peptone saline solution (0.1% m:v) and massaged for 1 minute to release the bacterial load. Aliquots of this solution were incubated in specific medium for bacterial growth for 48 hours at 37ºC for subsequent CFU counts. To evaluate the effects of disinfection on production, eggs previously disinfected by UV-C (9 min) and paraformaldehyde and NC eggs were candled between incubation days 10 and 13 and at the end of the incubation period to assess embryonic mortality. Hatchability distribution was performed every 8 hours. The 9 minutes 254nm UV-C light exposure was able to disinfect viable eggs and matched the effectiveness of the paraformaldehyde technique.(AU)


Objetivou-se avaliar a eficácia da luz ultravioleta na redução da carga bacteriana de cascas de ovos e o impacto na eclosão e na mortalidade embrionária observando-se a idade das matrizes (38, 42 e 48 semanas). Os ovos foram submetidos a diferentes períodos de exposição (cinco, sete e nove minutos) à luz UV (UV-C) com comprimento de onda de 254nm. Os controles foram ovos desinfetados com paraformaldeído (5,3g/m³) e ovos não desinfetados (NC). Após a desinfecção, os ovos foram colocados em sacos plásticos estéreis contendo 20mL de solução salina peptonada (0,1% m:v) e massageados por um (1) minuto para descolamento das bactérias. Alíquotas dessa solução foram incubadas em meio para crescimento bacteriano por 48 horas a 37ºC e contagem de UFC. Para avaliar os efeitos da desinfecção, ovos previamente desinfetados por UV-C (nove minutos) e ovos com paraformaldeído e NC foram submetidos à ovoscopia entre os dias 10 e 13 de incubação e ao final do período de incubação, para avaliação da mortalidade embrionária. A distribuição da eclodibilidade foi realizada a cada oito horas. A exposição à luz UV-C de 25nm de nove minutos desinfetou os ovos férteis e coincidiu com a eficácia do paraformaldeído.(AU)

ABSTRACT

Los embriones de tortugas marinas son susceptibles a perturbaciones ambientales, las cuales se traducen en anomalías en su fenotipo. Uno de los principales parámetros para la medición de estas perturbaciones es la asimetría en los carapachos (caparazones), que puede ser obtenida a través del Índice de Inestabilidad de Desarrollo (DIx, por sus siglas en inglés). La temperatura juega un rol importante, influenciando no solo en la futura supervivencia de los neonatos en temperaturas extremas (altas o bajas), sino en atributos específicos, tales como la determinación del sexo, o incluso puede ser la causa del incremento en el número de apéndices dérmicos (escudos) sobre la parte dorsal del carapacho. Este estudio evaluó la relación entre la temperatura de incubación con la asimetría de carapacho de los neonatos de Lepidochelys olivacea incubados en condiciones relocalizadas en el tortugario del Área de Usos Múltiples Hawaii en Santa Rosa, Guatemala. Para lograrlo, se replicaron las condiciones de anidación seleccionadas por las hembras anidadoras en nidos relocalizados, utilizando dispositivos termosensitivos para la toma de la temperatura durante el período de incubación. A través del cálculo del DIx en 210 neonatos, y mediante las pruebas de correlación de Kendall y Jockheere-Terpstra, se obtuvo que en valores de temperaturas extremas tanto altas (Kendall tau = .23, p < .001; JT = 2,891, p < .001), como bajas (Kendall tau u= -.21, p < .001; JT = 5,005, p < .001), así como en altos rangos de variación durante el período de desarrollo embrionario (Kendall tau = .23, p < .001; JT = 6,619, p < .05), los neonatos presentan una mayor asimetría.


Sea turtle embryos are susceptible to environmental disturbances, which result in abnormalities in their phenotype. One of the main parameters for measuring these disturbances is the asymmetry in the carapaces, which can be obtained through the Developmental Instability Index (DIx). Temperature plays an important role, influencing not only the future survival of hatchlings in extreme temperatures (high or low), but also specific attributes, such as sex determination, or it can even be the cause of the increase in the number of dermal appendages (shields) on the dorsal part of the carapace. This aimed to evaluate the relationship between incubation temperature and carapace asymmetry of Lepidochelys olivacea hatchlings incubated in relocated conditions in the Hawaii Multiple Use Area turtle rookery in Santa Rosa, Guatemala. To achieve this, the nesting conditions selected by the nesting females were replicated in relocated nests, using thermosensitive devices to measure the temperature during the incubation period. rough the calculation of DIx in 210 hatchlings, and using Kendall and Jockheere-Terpstra correlation tests, it was obtained that hatchlings present greater asymmetry in values of extreme temperatures, for both, high (Kendall: p<.05, tau = .232 ; JT p<.05, JT=2891) and low (Kendall: p<.05, tau= -.211 ; JT p<.05, JT=5005); as well as in high ranges of variation (Kendall: p<.05, tau= .231 ; JT p<.05, JT=6619), during the embryonic development period

ABSTRACT

This study aimed to evaluate the effects of different temperatures on incubation variables, performance, and morphometry of the duodenal mucosa of Japanese quails (Coturnix coturnix japonica) submitted to chronic heat stress after hatching. We distributed 540 eggs in three incubators with a temperature of 37.8°C and 60% of humidity. From the 6th day of incubation until hatching, the temperatures were adjusted to (37.8°C, 38.5°C and 39.5°C). After hatching, quails were evaluated for the quality score, weighed, and distributed in a completely randomized design with three incubation temperatures (37.8, 38.5, and 39.5°C) and two ambient temperatures (stress and thermoneutral). At 10, 20, 30, and 40 days they were weighed to determine the live weight (g) and weight gain(g). To collect the duodenum and determine morphometric parameters, we euthanized four quails of each treatment. The data were analyzed, and the differences between the means determined by the Tukey test at 5%. The incubation temperature of 39.5°C provided lower hatching rate and the live weight at birth; however, from the 10th day of age, increased live weight, weight gain, and positively influenced the morphological parameters of the duodenal mucosa in situations of chronic stress.(AU)


Objetivou-se avaliar os efeitos de diferentes temperaturas de incubação sobre as variáveis de incubação, desempenho e morfometria da mucosa duodenal de codornas japonesas (Coturnix coturnix japonica) submetidas ao estresse térmico crônico por calor após eclosão. Foram distribuídos 540 ovos em três incubadoras, com temperatura de 37,8°C e umidade 60%. A partir do sexto dia de incubação até a eclosão, as temperaturas foram ajustadas para 37,8°C, 38,5°C e 39,5°C. Após a eclosão, as codornas foram avaliadas quanto ao escore de qualidade, pesadas e distribuídas em um delineamento inteiramente ao acaso, com três temperaturas de incubação (37,8ºC, 38,5ºC e 39,5°C) e duas temperaturas ambientes (estresse e termoneutro). Aos 10, 20, 30 e 40 dias, foram pesadas para determinar o peso vivo (g) e o ganho de peso(g). Quatro codornas de cada tratamento foram eutanasiadas para coleta do duodeno, para determinar os parâmetros morfométricos. Os dados foram analisados e as diferenças entre as médias foram determinadas pelo teste de Tukey a 5%. A temperatura de incubação de 39,5°C proporcionou menor taxa de eclosão e menor peso vivo ao nascer, entretanto, a partir do 10° dia de idade, essa temperatura aumentou o peso vivo, o ganho de peso e influenciou positivamente os parâmetros morfológicos da mucosa duodenal em situações de estresse crônico.(AU)

Abstract Background: Preserving the genetic diversity of wild fish is an important consideration for restocking programs, as inbreeding can compromise progeny survival as well as impact the resilience of natural populations. Objective: To evaluate the influence of spawning method: semi-natural (SN) or strip-spawning (ST), and the number of breeders (1♀:3♂ and 2♀:6♂) on the reproductive efficiency and genetic diversity of B. orbignyanus progeny destined for restoration of wild stocks. Methods: Rates of fertilization, hatching and broodfish mortality were recorded. For genetic evaluations (allele frequency, observed and expected heterozygosity, Shannon index, inbreeding coefficient, molecular variance analysis, and genetic differentiation), breeders (n=24), and their progenies (90 larvae/treatment) were sampled and analyzed using eight microsatellite markers. Results: Higher fertilization and hatching rates, and lower broodfish mortality were observed for the SN method (p<0.05), whereas the number of breeders did not affect these parameters (p>0.05). Interaction between spawning method and number of breeders was not significant (p>0.05). The amplified microsatellite loci produced a total of 30 alleles, with sizes between 80 and 225 bp and their frequencies indicated an increase (p<0.05) of genetic diversity in the progenies, but low genetic differentiation between treatments (p>0.05). Conclusion: The spawning methods and number of breeders tested increased equally the genetic diversity of the progeny, with low genetic differentiation between treatments. In contrast, rates of fertilization, hatching and brood fish mortality revealed that the SN method resulted in the best reproductive efficiency due to the handling stress and injuries caused by ST. Thus, SN proves to be the most suitable spawning-method for B. orbignyanus in restocking programs.


Resumen Antecedentes: Mantener la diversidad genética de los peces salvajes es una consideración importante para los programas de repoblación, ya que la endogamia puede comprometer la supervivencia de la progenie y afectar la supervivencia de las poblaciones naturales. Objetivo: Evaluar la influencia del método de desove (seminatural - SN o en franjas - ST) y el número de reproductores (1♀:3♂ y 2♀:6♂) sobre la eficiencia reproductiva y la diversidad genética de progenies de B. orbignyanus destinados a la restauración de poblaciones silvestres. Métodos: Se monitorearon las tasas de fertilización, eclosión y mortalidad de reproductores. Para las evaluaciones genéticas (frecuencias alélicas, heterocigosidad observada y esperada, índice de Shannon, coeficiente de endogamia, análisis de varianza molecular y diferenciación genética) los reproductores (n=24) y su progenie (90 larvas/tratamiento) se muestrearon y analizaron utilizando ocho marcadores microsatélites. Resultados: La interacción entre el método de desove y el número de reproductores no fue significativa (p>0,05). Se obtuvieron mejores tasas de fecundación y eclosión (p<0,05), y una menor mortalidad de reproductores (p<0,05) con el método SN, mientras que el número de reproductores no tuvo efecto (p>0,05). Los loci de microsatélites amplificados produjeron un total de 30 alelos con tamaños entre 80 y 225 pb, y sus frecuencias indicaron un aumento (p<0,05) en la diversidad genética de las progenies, pero una baja diferenciación genética entre tratamientos (p>0,05). Conclusión: Los métodos de desove y el número de reproductores evaluados aumentaron de la misma manera la diversidad genética de las progenies, con baja diferenciación genética entre tratamientos. En contraste, las tasas de fecundación, eclosión y mortalidad de peces reproductores revelaron que el SN tuvo la mejor eficiencia reproductiva, un hecho relacionado con el estrés del manejo y las lesiones causadas por el ST. Por lo tanto, el SN demuestra ser el método de desove más adecuado para B. orbignyanus en los programas de repoblación.

ABSTRACT

Objective:To analyze the epidemiological characteristics, clinical characteristics and blood purification effect of mushroom poisoning in children.Methods:A retrospective study was conducted on 51 children with acute mushroom poisoning admitted to Hunan Children′s Hospital from 2002 to 2020.The epidemiological and clinical characteristics were analyzed, and the prognosis of children with different incubation periods was analyzed and compared.Among them, 36 critically ill children were treated with blood purification.Results:The age distribution was 66(43, 115)months.Mushroom poisoning had obvious seasonal and spatial aggregation.The first symptom was mainly manifested by digestive tract, which was manifested as vomiting, abdominal pain and diarrhea, accounting for 94.1% patients(48/51). The gastrointestinal tract type accounted for 45.1%(23/51) of the clinical types and the multi-organ damage type accounted for 51.0% patients(26/51). The length of hospital stay was 6(3, 11)days.Among them, early onset accounted for 45.1% patients(23/51), late onset accounted for 54.9% patients(28/51). Early onset hospitalization was shorter, about 4(2, 7)days, and fewer organs 1(0, 3) were damaged.The length of hospital stay of late onset was 8(3, 12)days, and the number of damaged organs was 4(2, 4). There was a statistically significant difference( P<0.05). Late onset patients had more severe organ function damage, including liver function, coagulation function, renal function, myocardial enzyme, and there was no significant difference in the final outcome between two groups.In 36 children who were given blood purification treatment, alanine aminotransferase, aspartate aminotransferase, prothrombin time, international normalized ratio, blood urea nitrogen, and lactate dehydrogenase were significantly improved, with statistically significant differences( P<0.05). Conclusion:Mushroom poisoning has obvious seasonal and geographical distribution characteristics; the first manifestation is mainly gastrointestinal symptoms; the clinical types are more common in gastrointestinal and multiple organ damage types.Children with early onset have shorter hospital stays and fewer complications than later onset.Blood purification treatment can significantly improve liver function, kidney function and blood coagulation function in children with toadstool poisoning.

ABSTRACT

Objective To investigate the disease progression and influencing factors of human immunodeficiency virus (HIV)-infected persons in Sanya area from 2010-2020. Methods Data of HIV infected cases reported in XX region from January 2014 to January 2020 were collected through the National AIDS Comprehensive Prevention and Control Information System. The incidence of AIDS was calculated by the life expectancy method, and the single factors influencing the progression of HIV infection were analyzed from the aspects of sex, age, BMI, nationality, marital status, educational level, highly active anti-retroviral therapy (HAART), anemia, route of transmission, CD4+ cell count, and co-infection of other viruses. Results A total of 761 HIV-infected cases were reported in Sanya areaFrom January 2010 to January 2020, of which 103 had progressed to AIDS (13.53%), the incidence of AIDS was 6.01/100 person-years, the average incubation period was 4.63 years; There were 31 death cases, and the all-cause mortality rate was 1.81/100 person years; Gender, age, HAART treatment status, anemia, CD4+ cell count, and other viral infections were single factors that affect the disease progression of HIV-infected persons (χ2=16.803, 13.362, 15.954, 20.119, 16.039, 15.711, P+ cell count, and other viral infections were independent risk factors that affect the disease progression of HIV-infected persons (P=0.014, 0.007, 0.017, 0.009, 0.001, 0.001). Conclusion The incubation period is an important stage of the disease progression of HIV-infected patients, moreover, age, HAART treatment status, anemia, CD4+ cell count, and other viral infections are all independent risk factors that affect the disease progression of HIV-infected patients.

BACKGROUND@#Current studies on the COVID-19 depicted a general incubation period distribution and did not examine whether the incubation period distribution varies across patients living in different geographical locations with varying environmental attributes. Profiling the incubation distributions geographically help to determine the appropriate quarantine duration for different regions.@*METHODS@#This retrospective study mainly applied big data analytics and methodology, using the publicly accessible clinical report for patients (n = 543) confirmed as infected in Shenzhen and Hefei, China. Based on 217 patients on whom the incubation period could be identified by the epidemiological method. Statistical and econometric methods were employed to investigate how the incubation distributions varied between infected cases reported in Shenzhen and Hefei.@*RESULTS@#The median incubation period of the COVID-19 for all the 217 infected patients was 8 days (95% CI 7 to 9), while median values were 9 days in Shenzhen and 4 days in Hefei. The incubation period probably has an inverse U-shaped association with the meteorological temperature. The warmer condition in the winter of Shenzhen, average environmental temperature between 10 °C to 15 °C, may decrease viral virulence and result in more extended incubation periods.@*CONCLUSION@#Case studies of the COVID-19 outbreak in Shenzhen and Hefei indicated that the incubation period of COVID-19 had exhibited evident geographical disparities, although the pathological causality between meteorological conditions and incubation period deserves further investigation. Methodologies based on big data released by local public health authorities are applicable for identifying incubation period and relevant epidemiological research.

Abstract The tegu (Salvator merianae) is the largest lizard found in Brazil and has a wide distribution, occurring in several biomes. The presence of fungi on reptilian eggs initially occurs on infertile eggs, which leads to a subsequent infection in fertile eggs, causing damage and death of embryos. The objective of this study was to identify the fungi that caused lesions in the eggs of tegu in captivity. Swabs were collected on infected and healthy eggshells, in an embryo that died and the substrate of the incubation box. Samples were submitted to saboraund dextrose agar. A total of 93.3% of positive samples were obtained for the fungi Fusarium (11/14), Trichoderma (1/15), Penicilium (1/15), Mucor (1/15), and yeasts of the genus Trichosporon. Therefore, Fusarium was the main agent that caused the lesion and death of embryos in the eggs of the tegu lizard. Sanitary control methods may be taken to avoid fungal infections in the eggs.


Resumo O teiú (Salvator merianae) é o maior lagarto encontrado no Brasil e possui ampla distribuição, sendo encontrado em vários biomas. A presença de fungos em ovos de répteis ocorre inicialmente em ovos inférteis, o que leva à uma infecção posterior em ovos férteis, gerando danos e morte dos embriões. O objetivo deste estudo foi o de identificar os fungos geradores de lesões em ovos de teiú em cativeiro. Foram coletados suabes na casca de ovos infectados e sadios, além de um embrião que veio a óbito, e do substrato presente na caixa de incubação. As amostras foram submetidas ao cultivo em ágar Saboraund dextrose. Um total de 93,3% de amostras foram positivas para os fungos Fusarium (11/15), Trichoderma (1/15), Penicillium (1/15), Mucor (1/15) e leveduras do gênero Trichosporon (3/15). Logo, Fusarium foi o principal agente causador das lesões e morte de embriões nos ovos do lagarto teiú. Métodos de controle sanitário podem ser adotados para evitar infecção fúngicas nos ovos.

Objective@#To study the early dynamics of the epidemic of coronavirus disease (COVID-19) in China from 15 to 31 January, 2020, and estimate the corresponding epidemiological parameters (incubation period, generation interval and basic reproduction number) of the epidemic.@*Methods@#By means of Weibull, Gamma and Lognormal distributions methods, we estimated the probability distribution of the incubation period and generation interval data obtained from the reported COVID-19 cases. Moreover, the AIC criterion was used to determine the optimal distribution. Considering the epidemic is ongoing, the exponential growth model was used to fit the incidence data of COVID-19 from 10 to 31 January, 2020, and exponential growth method, maximum likelihood method and SEIR model were used to estimate the basic reproduction number.@*Results@#Early COVID-19 cases kept an increase in exponential growth manner before 26 January, 2020, then the increase trend became slower. The average incubation period was 5.01 (95%CI: 4.31-5.69) days; the average generation interval was 6.03 (95%CI: 5.20-6.91) days. The basic reproduction number was estimated to be 3.74 (95%CI: 3.63-3.87), 3.16 (95%CI: 2.90-3.43), and 3.91 (95%CI: 3.71-4.11) by three methods, respectively.@*Conclusions@#The Gamma distribution fits both the generation interval and incubation period best, and the mean value of generation interval is 1.02 day longer than that of incubation period. The relatively high basic reproduction number indicates that the epidemic is still serious; Based on our analysis, the turning point of the epidemic would be seen on 26 January, the growth rate would be lower afterwards.

Since December 2019, Corona Virus Disease (COVID-19), a new emerging infection disease occurred in Wuhan, has spread in 27 countries and regions. The clusters of many cases were reported with the epidemic progresses. We collected currently available information for 377 COVID-19 clusters (1 719 cases), excluded the hospital clusters and Hubei cases, during the period from January 1, 2020 to February 20, 2020. There were 297 family clusters (79%), case median 4; 39 clusters of dining (10%), case median 5; 23 clusters of shopping malls or supermarkets (6%), case median 13; 12 clusters of work units (3%), case median 6, and 6 clusters of transportation. We selected 325 cases to estimate the incubation period and found its range is 1 to 20 days, median was 7 days, and mode was 4 days. The analysis of the epidemic situation in a department store in China indicates that there is a possibility of patients as the source of infection during the incubation period of the epidemic. From February 5, 2020 to February 21, 2020, 634 persons were infected in the Diamond Princess Liner. All persons are susceptible to SARS-CoV-2. The older, patients during the incubation period and the worse environment may be the cause of the cases rising. The progress of the two typical outbreaks clearly demonstrates the spread of the early cases in Wuhan. Whatever happens, screening and isolating close contacts remains essential except for clinical treatment during the epidemic. Especially for the healthy people in the epidemic area, isolation is the key.
